Metabolic processes usually ensure that heat is released. What could be more obvious than to consider heat as an indicator of life processes – whether in humans, animals, plants or microorganisms? As heat is so closely linked to metabolism, it indicates its changes at the moment they occur. The heat output therefore contains important information about the speed and material flows of biological conversions. These are ideal prerequisites for controlling difficult bioprocesses, such as the microbiological conversion of toxic pollutants into valuable substances. Biotechnological processes are finding their way into the pharmaceutical, agricultural and chemical industries with increasing frequency and speed. If traditional synthesis steps in the manufacture of medicines and chemical products can be replaced by biotechnological processes, production costs and environmental pollution can be reduced. This is why there is great interest in monitoring, controlling and analysing bioprocesses more precisely.
Calorimeters or biocalorimeters, which measure the heat release as a function of time and thus provide an exact picture of the metabolism of microbiological activities, are an essential tool for this. Like the smaller model Calmetrix Biocal 2000 with two measuring positions, the isothermal four-channel biocalorimeter Calmetrix Biocal 4000 is used here. The biocalorimeter operates isothermally in the temperature range 5°C to 70°C, has four thermally decoupled measuring positions (no cross-talk visible), each of which is coupled with a variable reference in twin design and, with a sample volume of up to 125 ml each, offers the user enough leeway to be able to precisely analyse even less active processes in the lower microwatt range using an appropriate sample volume. The sample vessels used are plastic beakers with lids (disposable) or stainless steel vessels with plastic lids (sterilisable). The intuitive software is used to store the test data and a few measurement parameters. Once started, the heat development is recorded online. The measurement data thus provides both qualitative and quantitative information on the time course and extent of the reactions taking place. The respective “reference sample” consists of several metal discs, mounted on a holder, and can be quickly and variably adapted to the sample volume or its mass in terms of thermal mass.
If more measuring channels are required for a higher sample throughput and smaller sample volumes (20 ml) are sufficient or the available sample volume needs to be even larger (up to 450 ml), the Calmetrix I-Cal Flex isothermal multi-channel calorimeter can be a good alternative.
